<dc:title>2022–2023 DNA Barcode Dataset of Fishes from Dongting Lake (COI and 12S rRNA)</dc:title>

<dc:description>This dataset is a DNA‑barcode reference dataset for fishes of Dongting Lake based on two mitochondrial molecular markers. Samples were collected from 16 monitoring sites across Dongting Lake during 2022‑2023, covering typical habitats including inflowing rivers, lake‑center areas and littoral zones. A total of 229 COI sequences and 243 12S rRNA sequences were generated, representing 75 fish species belonging to 46 genera and 15 families, plus two hybrid groups (hybrid Carassius and hybrid Acipenser). Fish specimens were morphologically identified in the field; fin‑clip tissues were used for DNA extraction. Target fragments were amplified with primer pairs Fish‑F1/Fish‑R1 for COI and AcMDB07‑F/AcMDB07‑R for 12S rRNA, followed by bidirectional Sanger sequencing. Raw sequences were quality‑filtered, assembled and trimmed using SeqMan Pro and MEGA 11.0. Intra‑ and inter‑specific genetic distances were calculated under the Kimura‑2‑parameter (K2P) model, and Neighbor‑Joining (NJ) phylogenetic trees were constructed. All COI sequences retained the standard 655 bp barcode fragment compliant with BOLD specifications. The dataset comprises sequence files, sample‑collection metadata, genetic‑distance statistics and phylogenetic‑tree outputs. It supplements missing reference barcodes for local Dongting Lake fishes in public databases, providing barcode data for eight species absent from public repositories. This dataset supports eDNA metabarcoding taxonomic assignment, freshwater fish species identification, fish‑biodiversity assessment and evaluation of the ten‑year fishing ban in the middle‑lower Yangtze River, and provides molecular baseline data for watershed‑scale biodiversity conservation. This dataset corresponds to the paper “Construction of a DNA Barcode Database for Fishes in Dongting Lake Based on Two Mitochondrial Molecular Markers”.</dc:description>
