<dc:description>After extracting tumor tissues from LLC tumor bearing Ctrl, Batf3-/-, and Xcr1CreTrim33f/f mice, single-cell suspensions were prepared. Cells undergo CD16/32 blockade and flow cytometry antibody staining: FVD506, CD45, CD3, NK1.1, CD19, CD8, CD4. Machine sorting CD45+CD3+CD8+T cells. After sorting, centrifuge the cells, discard the supernatant, add Trizol to extract RNA, and deliver it to BGI Genomics for subsequent work. Library construction: Total RNA was processed using mRNA enrichment method. mRNA enrichment: PolyA tail mRNA was enriched using magnetic beads with OligodT. Use interrupt buffer to fragment the obtained RNA, perform reverse transcription with random N6 primers, and then synthesize cDNA double stranded DNA to form double stranded DNA. Flatten the ends of the synthesized double stranded DNA and phosphorylate the 5 'end to form a sticky end with a protruding "A" at the 3' end, and then connect it to a bubbly junction with a protruding "T" at the 3 'end. The connecting product is amplified by PCR using specific primers. The PCR product undergoes thermal denaturation to form a single stranded DNA, which is then cyclized using a bridge primer to obtain a single stranded circular DNA library. After the constructed library passes the quality inspection, it will be sequenced. Using DNBSEQ platform and PE150 (read length) sequencing. Subsequent use of Dr. Tom's multi omics data mining system（ https://biosys.bgi.com ）Perform data analysis, plotting, and mining. </dc:description>
