<dc:title>pH-Responsive EF-TK-GNTs Nanoplatform Orchestrates Chemo-Photothermal Therapy of Osteosarcoma via ERK/mTOR Axis</dc:title>

<dc:description>Osteosarcoma is the predominant primary bone malignancy in adolescents, and the survival of patients with metastases has remained stagnant for over three decades. Here, we describe a rationally designed, pH- and NIR-dual-responsive nanotherapeutic system, EF-TK-GNTs, in which ethyl ferulate (EF) is conjugated to gold nanotriangles (GNTs) via a thioketal (TK) linker. This architecture allowed acidic pH-triggered EF release, whereas 808 nm near-infrared (NIR) light elicited a strong hyperthermic response (ΔT > 50 °C). In K7M2 murine osteosarcoma cells, the chemo-photothermal combination (G4) produced a pronounced combined cytotoxic effect (~74.2% reduction in viability), migration inhibition, and apoptosis. Mechanistically, we observed that 808 nm-induced photothermal stress was accompanied by marked HSP70 upregulation, which may act as a negative regulator contributing to ERK suppression, while the released EF inhibited mTOR activity. This dual attenuation of two key pro-survival pathways was associated with disruption of cellular homeostasis and commitment to mitochondrial apoptosis in vitro. These findings suggest that EF-TK-GNTs may serve as a promising chemo-photothermal platform that attenuates ERK-mTOR co-signaling in osteosarcoma cells, providing an in vitro proof-of-concept for further preclinical validation.</dc:description>
