<dc:title>Identification of Sox9a/Sox9b Genes in Nibea albiflora Based on tblastn Alignment</dc:title>

<dc:description>Supplementary material of "Molecular Characteristics of Sox9a Gene and Its Expression Regulation on dmrt1a in yellow drum" (Manuscript No. SSSWXB-2025-0462. R2): To determine whether the cloned sox9 gene belonged to sox9a or sox9b, the amino acid sequences of Sox9a and Sox9b from the closely related species Larimichthys crocea were selected. A tblastn search was performed on the NCBI database with the species restricted to Nibea albiflora. The results showed that L. crocea sox9a shared the highest sequence identity with the sequence on chromosome 18 of N. albiflora (Supplementary material 1), while sox9b exhibited the highest identity with the sequence on chromosome 14 of N. albiflora (Supplementary material 2), indicating that both sox9a and sox9b exist in N. albiflora. Afterwards, the cloned Sox9 amino acid sequence was aligned against the N. albiflora genome via NCBI tblastn. The matched gene fragment was located on chromosome 18 (Supplementary material 3), confirming that the sox9 gene cloned in this study is sox9a.</dc:description>
