<dc:title>Construction of Silver Nanoparticle-based Artificial Hydrolases via Conformational Engineering and Study of Its Catalytic Mechanism</dc:title>
  <dc:identifier>doi:10.57760/sciencedb.37697</dc:identifier>
  <dc:language>en</dc:language>
<dc:description>This dataset originates from a study on the construction of a highly active silver-based artificial hydrolase (Silverzyme) based on conformational engineering strategies. Using silver nanoparticles (AgNPs) as the scaffold, the study successfully mimicked the catalytic center of &amp;alpha;-chymotrypsin&amp;mdash;including the catalytic triad, oxyanion hole, and substrate-binding site&amp;mdash;through peptide design and a three-step conformational control method (induction of &amp;alpha;-helical conformation by trifluoroethanol, stabilization of the conformation via Ag&amp;ndash;S bonds, and removal of the inducer by freeze-drying). The dataset primarily includes the following content:Circular Dichroism (CD) data: Used to demonstrate the formation and maintenance of the &amp;alpha;-helical conformation of the peptides on the AgNP surface, thereby validating the successful implementation of conformational engineering.Stopped-flow kinetics data: Including comparisons of catalytic activity before and after mutation of key residues, used to demonstrate the functional necessity of the catalytic triad, oxyanion hole, and substrate-binding site, as well as their synergistic effects.Catalytic activity data: The turnover number per active site measured using p-nitrophenyl acetate (p-NPA) as the substrate.Substrate scope expansion data: Activity data showing the catalytic degradation of diethyl phthalate (DEP), an environmental priority pollutant, by Silverzyme, demonstrating its ability to hydrolyze non-activated esters.</dc:description>
