<dc:title>Microbial community assembly in rapeseed under uranium stress: 16S rRNA sequencing across soil-root-leaf niches</dc:title>
  <dc:identifier>doi:10.57760/sciencedb.43243</dc:identifier>
  <dc:language>en</dc:language>
<dc:description>This dataset comprises 16S rRNA gene amplicon sequencing data generated from a pot experiment investigating microbial community dynamics under uranium stress in rapeseed (Brassica napus). Soil samples were collected from three treatments: original soil, sterilized soil, and sterilized-inoculated soil, with or without 150 mg kg⁻&amp;sup1; uranium addition. Four ecological niches were sampled: bulk soil, rhizosphere soil, root endosphere, and leaf phyllosphere. The V3&amp;ndash;V4 region of the 16S rRNA gene was amplified using primers 338F/806R and sequenced on the Illumina MiSeq platform (2&amp;times;300 bp paired-end). Raw reads were quality-filtered, denoised, and merged using the DADA2 pipeline, and operational taxonomic units (OTUs) were clustered at 97% similarity. Taxonomic assignment was performed against the Greengenes 13.8 database. Quality control included negative controls (extraction blanks and PCR negatives) and rarefaction to 95% of the minimum sample depth to ensure comparability. These data are valuable for studying microbial community assembly, niche differentiation, functional redundancy/compensation, and plant&amp;ndash;microbe interactions in uranium-contaminated soils, and can support comparative analyses with other heavy-metal or phytoremediation studies.
