2026-09-11 Construction of single-cell Raman reference phenotypes of type I and type II methanotrophs under 13CH4 labeling doi:10.57760/sciencedb.cjae.000a2 en This dataset provides experimental source data associated with manuscript 2026-0673 submitted to the Chinese Journal of Applied Ecology. The study used four type I and four type II methanotrophic strains in pure culture. Single-cell Raman spectra were collected under ¹²CH₄ control and ¹³CH₄-labeling treatments, and the effects of growth stage on spectral characteristics were examined. The shared files are organized into four components: single-cell Raman spectra under the ¹²CH₄ treatment; single-cell Raman spectra under ¹³CH₄ labeling; single-cell Raman spectra of Methylomonas sp. LW13 at five growth stages; and growth and methane utilization data for the representative strains LW13 and SD4, including OD₆₀₀ and CH₄ concentration measurements. These data support analyses of isotope-labeling-associated spectral changes, growth-stage-related spectral characteristics, and spectral comparisons between type I and type II reference strains.
