<dc:title>Dataset on In vitro anti-photoaging and whitening activities of different extracts from the whole plant of Paeonia suffruticosa</dc:title>

<dc:description>This dataset focuses on the evaluation of anti-photoaging and skin-whitening bioactivities of extracts from different whole-plant parts of tree peonies and their compound formulations. It covers the complete experimental workflow from sample preparation to functional bioactivity validation, and can provide systematic basic data support for the screening of natural active components from tree peonies and the development of cosmetic and edible raw materials.Tree peony petals, leaves, root bark and seed coats used in the experiments were all collected from the standardized tree peony planting base in Luoyang City, Henan Province, in April and September 2024. Three independent biological replicates were set for each plant part. Fresh samples were processed to prepare extracts for subsequent bioactivity assays. For functional bioactivity evaluation, a UVB-induced photodamage model of HaCaT cells and a B16 melanoma cell model were constructed separately. A series of indicators were detected sequentially, including cell viability, transcription levels of senescence-related genes, antioxidant-related genes, intracellular reactive oxygen species (ROS) levels, melanin content, intracellular tyrosinase activity, and transcription levels of whitening-related genes. The major equipment adopted in the experiments included a CO₂ cell incubator, microplate reader, high-speed refrigerated centrifuge, inverted fluorescence microscope, and UV irradiator. The dataset contains a total of 18 independent data files, detailed as follows:Supplement Table 1 contains data on the quantitative detection of polyphenols, peptides, flavonoids and paeonol (row labels correspond to the names of each detection indicator, and column labels correspond to the serial numbers of biological replicates);Figure 2: Detection data of HaCaT cell viability;Figure 3: Detection data of HaCaT cell viability;Figure 4: Detection data on transcription levels of senescence-related genes in HaCaT cells;Figure 5: Detection data on transcription levels of antioxidant-related genes in HaCaT cells;Figure 6: Detection data on intracellular ROS levels in HaCaT cells;Figure 7: Detection data of B16 cell viability;Figure 8: Detection data on melanin production in B16 cells;Figure 9: Detection data on intracellular tyrosinase activity in B16 cells;Figure 10: Detection data on transcription levels of whitening-related genes in B16 cells;Figure 11A: Data for safe concentration detection of MP on HaCaT cells;Figure 11B: Data for detecting the protective effect of MP on the viability of UVB-damaged HaCaT cells;Figure 11C–G: Detection data of senescence-related genes in MP-treated HaCaT cells;Figure 11H–I: Detection data of antioxidant-related genes in MP-treated HaCaT cells;Figure 12A: Data for safe concentration detection of MP on B16 cells;Figure 12B: Detection data on melanin content in MP-treated B16 cells;Figure 12C: Detection data on intracellular tyrosinase activity in MP-treated B16 cells;Figure 12D–G: Detection data of whitening-related genes in MP-treated B16 cells.(Row labels correspond to different experimental groups, and column labels correspond to biological replicate numbers and data descriptions.)The measurement units involved include μg/mL (administration concentration) and % (cell viability, melanin content, tyrosinase activity). No systematic data loss exists in this dataset. Data errors mainly stem from biological individual differences of samples and fluctuation of instrument detection systems. The intra-group coefficient of variation (CV) of all cellular activity indicators was controlled within 10%. All experiments were performed with at least three biological replicates and three technical replicates, and all results are presented as mean ± standard deviation, which effectively reduces the influence of random errors on data reliability. All experimental operations for this dataset strictly complied with corresponding experimental specifications and instrument operating standards, with complete and traceable data records. The dataset can be directly applied to subsequent comparative analysis of natural product bioactivities, in-depth exploration of action mechanisms, and reference for the development of cosmetic raw materials.</dc:description>
