<dc:description>•This study aims to investigate the effect of Atractylodis Macrocephalae Rhizoma - Atractylodis Rhizoma (AM-AL) on liver autophagy in ob/ob obese mice through the PI3K/AKT/FOXO1 pathway. Male C57BL/6J mice served as the normal group, while male ob/ob mice were divided into the model group, metformin group (70 mg/kg), and AM-AL low dose groups (6 g/kg) and high-dose groups (12 g/kg), with six mice per group, all fed regular chow. The normal and model groups were gavaged with the same volume of saline, twice daily, for 10 consecutive weeks. Serum total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), aspartate aminotransferase (AST), and alanine aminotransferase (ALT) levels were measured using a fully automated biochemical analyzer; liver pathology was observed by HE staining and Oil Red O staining; liver inflammation markers interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were detected by ELISA; Western blot was used to assess the protein expression levels of liver autophagy markers ATG7, Beclin1, LC3Ⅱ/Ⅰ, P62, and the proteins PI3K, AKT, and FOXO1. The results show that AM-AL-H group reduced liver weight; decreased HDL-C and LDL-C levels; decreased TNF-α levels; increased Beclin1 protein expression; reduced LC3Ⅱ/Ⅰ and P62 protein expression; and increased PI3K, AKT and FOXO1 protein expression. The above results indicate that AM-AL can promote liver autophagy, possibly through regulation of the PI3K/AKT/FOXO1 signaling pathway.</dc:description>
