<dc:title>Transcriptomic profiling of midbrain tissue in mice with acute diquat poisoning</dc:title>

<dc:description>Objective Acute diquat poisoning can cause severe central nervous system injury and multiple organ dysfunction, yet the underlying molecular pathological mechanisms remain unclear. This study employed reference-based transcriptome sequencing to analyze global gene expression profile changes in acute diquat-poisoned mice, aiming to elucidate the toxicological mechanisms of diquat and identify potential therapeutic targets. Methods Adult C57BL/6 mice were selected; the diquat‑treated group received 300 mg/kg diquat by oral gavage, while the control group received an equal volume of saline. On day 24 after exposure, the mice were euthanized, and total RNA was isolated from midbrain tissues. Transcriptome sequencing was performed on an Illumina NovaSeq 6000 platform. Raw sequencing data underwent rigorous quality control, were aligned to the reference genome, and differentially expressed genes were identified using DESeq2. Subsequently, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis and protein‑protein interaction network analysis were conducted for multi‑level functional annotation of candidate hub genes. Results KEGG enrichment revealed that upregulated genes were mainly enriched in pathways, including apoptosis, TNF signaling pathway, AMPK and HIF‑1 signaling pathways, and glycolytic reprogramming in cancer. Downregulated genes were also predominantly enriched in pathways, including neuroactive ligand‑receptor interaction, glutamatergic postsynaptic components, axon guidance, cell adhesion molecules, Ras signaling, and various types of lipid biosynthesis pathways. Moreover, Tnf, Vegfa, Gli1, Nup214, and Xpo1 were identified as important upregulated hub genes. Conclusions This study preliminarily reveals the transcriptomic signatures of acute diquat toxicity, which will facilitate future interventional experiments targeting these candidates and provide a scientific basis for such investigations.</dc:description>
